The movie, Maa Behen follows a widow, 'Rekha' (played by Madhuri Dixit), who lives with her daughter, 'Sushma' (played by Dharna Durga). Her elder daughter, 'Jaya' (played by Triptii Dimri) is married and lives with her husband. 'Rekha' faces a lot of judgement in her neighbourhood as she refuses to follow the traditional things expected from a woman.

'Rekha's' daughter, 'Jaya' is in a very unhealthy marriage as she does not receives the respect she deserves. She is even blamed for not having children. On the other hand, 'Sushma' is trying to build her life. The movie picks up pace when 'Gupta Ji' (played by Ravi Kishan) enters. He is 'Rekha's' neighbour, who has very wrong intentions. One night he goes to 'Rekha's' house, and tries to behave inappropriately. In her defense, 'Rekha' hits him, leading him to lose consciousness.

The incident scares 'Rekha' and her family and they do not want 'Gupta Ji' to tell what happened to anybody. So 'Rekha' and her daughters decide to hide him temporarily. Soon, this turns into a case of kidnapping. Eventually, when the neighbourhood notices that 'Gupta Ji' is missing, they try to find him, because of which 'Rekha' is forced to lie.

Later, the situation worsens, when 'Gupta Ji's' family receives a ransom call, leaving 'Rekha' confused as she did not call them. During this time, one of the most powerful moments of the film takes place, when 'Jaya' decides to confront her husband regarding how his family treats and disrespects her. In the heated confrontation, 'Jaya' takes a stand for herself and demands a divorce.

Amid all the chaos, ‘Gupta Ji’ somehow manages to escape. And eventually, when he gets to know about who actually kidnapped him, he plans to take revenge; however, 'Rekha' and her daughters were not ready to stay silent. The women decide to arrange a meeting with 'Gupta Ji' and record his unacceptable behaviour. When he reaches, he shows his true colours. Later, when he realises that he was recorded, he tries to threaten the women. But now the women are not scared. Finally, ‘Gupta Ji’ faces the consequences of his actions.

While it seems like everything ended on a good note, one question remains the same. Well, towards the end, the movie reveals that the person to demand the ransom was ‘Gupta Ji’s’ daughter, ‘Goldy’, who was being forced by her parents to get married. When her father got kidnapped, she saw it as the perfect opportunity to escape.
She pretended to be the kidnapper and demanded around five lakh rupees. And with that money she ran away from her house and started a new life. On the other hand, ‘Rekha’, ‘Sushma’ and ‘Jaya’ also get the end they craved for. 'Rekha' finally stops the society from judging her, 'Jaya' walks out from an unhappy marriage and 'Sushma' continues to follow her passion. Lastly, ‘Gupta Ji’ has lost all his power that he imposed on other women.
