To give a brief about her professional life, Rupali made her television debut with the show, Sukanya but rose to prominence with her negative role in Sanjivani. Rupali has beautifully managed her personal and professional life.

It was on February 6, 2013, when Rupali Ganguly and Ashwin K Verma had tied the knot in an intimate wedding and two years later, the couple welcomed their first child, Rudransh into their world. For Rupali, becoming a mother was an ambition but her pregnancy didn't come easy as she had trouble conceiving her son. Due to thyroid issues, Rupali's fertility count decreased but despite difficulties, she gave birth to her son, whom she calls a 'miracle'.

When Rudransh turned 5 on August 25, 2020, Rupali Ganguly had posted a heartfelt note for her 'biggest blessing' and 'miracle'. Sharing an array of Rudransh's pictures, Rupali had written, ""A child gives birth to a mother" Thank you Rudransh for choosing me to be your Maa. Happy Birthday my baby my jaan… I couldn't have asked for a better miracle from Matarani. My biggest blessing. May u keep making ur family proud and the world a better place my child." After giving birth to Rudransh, Rupali Ganguly had gained 28 kgs and was body-shamed by women around her. For a few days, Rupali had not even stepped out of her house due to the fear of being judged for the post-pregnancy weight gain. Though the second season of Sarabhai v/s Sarabhai helped Rupali to step out but she realised she didn't want to leave her child. If not for Anupamaa, Rupali would have extended her sabbatical.
Rupali Ganguly and Ashwin K Verma had completed eight years of marital bliss on February 6, 2021, and to mark the same, she had posted their selfie. Rupali had captioned it as "My gummy smile when I truelyyyy happy. May this happiness always remain for both of us. May God give you the strength to be with me forever and after. Love u Patidev. Happy 8 years. THU THU THU."
